Yea they work together, but be aware of the differences of multiple GPUs in DirectX 12 vs DirectX 11, the gains are not as dramatic, so you will be better off waiting for Vega 10.

Unless you are trying to render at 7680x4320 one RX 480 should be OK for general gaming. Dual cards are OK but more are not really going to get you very far.

Z170 does not have enough PCI Express lanes for more than 2 video cards

one more question, if i change GPU with

ASUS Radeon RX 480 8GB, it wont give real difference right ?

It's the same card by a better manufacturer, so there will be no real difference.
